The sample is measured for volume (at RVH Lab), and then sent to Mayo to be analyzed microscopically to determine the number of sperm present, the number of moving or motile sperm, and the properties of the sperm motility.

Requirements Preps

Abstain from ejaculation for a minimum of 2, but not longer than 7 days, before collecting the specimen, unless the physician specifies otherwise. Do not use lubricants as many lubricants can kill sperm. Avoid vaginal and oral secretions which will contaminate your specimen. Sample should be obtained by masturbation and ejaculated into a wide-mouthed sterile collection container. Transport to outpatient laboratory within 1 hour of collection. During transportation to laboratory, sample should be kept at ambient temperature (close to body). Patient to proceed directly to outpatient laboratory to drop off specimen. Then patient is to proceed to registration after specimen is delivered to outpatient laboratory.

Clinical Use

Semen is composed of spermatozoa suspended in seminal fluid (plasma). The function of the seminal fluid is to provide nutrition and volume for conveying the spermatozoa to the endocervical mucus.

Male infertility can be affected by a number of causes. Chief among these is a decrease in the number of viable sperm. Other causes include sperm with abnormal morphology and abnormalities of the seminal fluid.
